I am unable to change the difficulty setting within the game, even though I have done it earlier in the game. Any ideas why and a solution? Thanks.

You can only lower the difficulty after starting the game sadly, it won't allow you to raise it back up again. This is not a glitch it tells you this when choosing your difficulty before starting the game.

That may only be true of the achievement-based difficulties. I can switch between Casual and Normal without a problem, even during boss fights.
